چکیده

Novel immunomagnetic microspheres have been prepared for separation of CD4 + lymphocytes. The magnetic nanoparticles with a diameter of approximately 78 nm were first synthesized by co-precipitation from ferrous and ferric iron solutions and subsequently entrapped within poly (glycidyl methacrylate) (PGMA) microspheres by a swelling and penetration process. Monoclonal antibody specific to CD4 molecules expressed on CD4 + lymphocytes was conjugated to the surface of magnetic PGMA microspheres through covalent bonding between epoxide functional groups on the microspheres surface and primary amine groups of the antibodies. Flow cytometer was used to confirm the presence of antibody on the surface of microspheres. The new immunomagnetic microspheres are promising to be used in a various biomedical applications including diagnosis, and monitoring of human diseases.

Preparation of Novel Poly(hydroxyethyl methacrylate-coglycidyl methacrylate)-Grafted Core-Shell Magnetic Chitosan Microspheres and Immobilization of Lactase

Poly(hydroxyethyl methacrylate-co-glycidyl methacrylate)-grafted magnetic chitosan microspheres (HG-MCM) were prepared using reversed-phase suspension polymerization method. The HG-MCM presented a core-shell structure and regular spherical shape with poly(hydroxyethyl methacrylate-co-glycidyl methacrylate) grafted onto the chitosan layer coating the Fe3O4 cores. The average diameter of the magn...

Introduction: Undoubtedly, reproduction is a necessity for survival and successful pregnancy is an immunological paradox. In the present study, we investigated the proportional changes of CD4+CD25bright T cells, CD4+CD25dim T cells in peripheral blood in unexplained recurrent spontaneous abortions (URSA) and compared it with normal pregnant women by antibody monoclonal method. Methods: The st...

Magnetic poly(2-hydroxyethyl methacrylate) microspheres for affinity purification of monospecific anti-p46 kDa/Myo1C antibodies for early diagnosis of multiple sclerosis patients

The aim of the present study is to develop new magnetic polymer microspheres with functional groups available for easy protein and antibody binding. Monodisperse macroporous poly(2-hydroxyethyl methacrylate) (PHEMA-COOH) microspheres ~4 µm in diameter and containing ∼1 mmol COOH/g were synthesized by multistep swelling polymerization of 2-hydroxyethyl methacrylate (HEMA), ethylene dimethacrylat...

PEGylation of magnetic poly(glycidyl methacrylate) microparticles for microfluidic bioassays.

In this study, magnetic poly(glycidyl methacrylate) microparticles containing carboxyl groups (PGMA-COOH) were coated using highly hydrophilic polymer poly(ethylene glycol) (PEG). PEG was used to reduce nonspecific interactions with proteins and cells while decreasing adhesion of particles to the walls of a microfluidic devices from poly(dimethylsiloxane) (PDMS) and cyclic olefin copolymer (COC...
